Rapid Lentivirus Titration Cassette ( LentiTitre) is a lentivirus titration kit for rapid and accurate estimation of the functional lentivirus titer in less than 10 minutes. LentiTitre is a lateral flow immunoassay (LFIA) based titration device, based on p24 detection with sensitive gold-conjugated antibody. Lentiviral supernatant standards functionally titrated at 106 TU/mL are included in the kit. By comparing the intensities of the test lines of the standards and that of unknown sample, the cassette can help accurately estimate lentiviral TU titer, using p24 as a gauge. It can also be used to determine the optimal time when packaging preparation is ready for harvesting.

Protocol

• Prepare the standards: Use the 10^6 TU/ml stock directly from the provided vial. Make two additional standards by diluting with cell culture medium to desired titers. Suggest 10-fold dilutions to 10^5 TU/ml and 10^4 TU/ml, or 5-fold dilutions to 2x10^5 TU/ml and 4x10^4 TU/ml.
• Place the cassette horizontally.
• Add 100 µL of each standard to one of the sample wells.
• Add 100 µL of packaging supernatant to one of the sample wells.
• Wait for 10 minutes
• The Control line will always appear. Determine the titer of unknown lentiviral preparation by comparing the intensity of its Test line against those of the standards.
